Lack of NLRP3 inflammasome activation reduces age-dependent sarcopenia and mitochondrial dysfunction, favoring the prophylactic effect of melatonin
To investigate the role of NLRP3 inflammasome in muscular aging, we evaluated here the morphological and functional markers of sarcopenia in the NLRP3-knockout mice, as well as the beneficial effect of melatonin supplementation. The gastrocnemius muscles of young (3 months), early-aged (12 months), and old-aged (24 months) NLRP3-knockout female mice were examined. Moreover, locomotor activity and apoptosis were assessed. The ... Read more

Melatonin, clock genes and mitochondria in sepsis
After the characterization of the central pacemaker in the suprachiasmatic nucleus, the expression of clock genes was identified in several peripheral tissues including the immune system. The hierarchical control from the central clock to peripheral clocks extends to other functions including endocrine, metabolic, immune, and mitochondrial responses. Increasing evidence links the disruption of the clock genes expression with multiple diseases ... Read more

Identification of morphological markers of sarcopenia at early stage of aging in skeletal muscle of mice
The gastrocnemiusmuscle (GM) of young (3months) and aged (12 months) femalewild-type C57/BL6 micewas examined by light and electron microscopy, looking for the presence of structural changes at early stage of the aging process.Morphometrical parameters including body and gastrocnemius weights, number and type ofmuscle fibers, cross section area (CSA), perimeter, and Feret's diameter of single muscle fiber, were measured. Moreover, lengths of the ... NLRP3 inflammasome affects mitochondrial morphology in skeletal muscle of mice
NLRP3 inflammasome seems to affect negatively skeletal muscle and mitochondria morphology. NLRP3 is associated with mitochondrial dysfunction and damage after its activation by inflammatory signals. Finally, these changes may reflect loss of efficacy of the skeletal muscle strength. Read more
